当外界环境发生变化后植物能够改变自身功能性状及时调整适应策略, 因此植物功能性状能够有效地反映植物对草地利用变化的响应, 然而在内蒙古草原从植物功能性状角度开展草地利用方式影响的研究略少。该研究以内蒙古典型草原大针茅(Stipa grandis)、羊草(Leymus chinensis)、糙隐子草(Cleistogenes squarrosa)和冷蒿(Artemisia frigida) 4种主要优势种为研究对象, 探讨在长期自由放牧、割草、短期围封和长期无干扰的影响下优势种植物功能性状的差异, 以期从功能性状视角, 揭示植物在受到外界干扰后的适应策略, 旨为天然草地的可持续管理提供基础数据支持和科学依据。结果表明: 1)除糙隐子草外, 在长期放牧后内蒙古典型草原优势植物植株高度、根长和植物碳氮含量降低, 这些性状的变化能够使植物个体小型化, 适口性降低, 表明植物通过逃避放牧的策略适应长期自由放牧的干扰; 在割草管理方式下, 优势种的高度和比叶面积有增加的趋势, 其中冷蒿的氮含量对割草响应最敏感, 其根、茎、叶中的氮含量均在割草样地最低; 围封和长期无干扰处理下植物的碳氮含量增加, 表明在干扰强度降低后, 植物通过功能性状的改变从资源获取策略向资源储藏策略转变。2)对优势种功能性状集合分析表明, 糙隐子草具有较低的植株高度和较高的比叶面积, 冷蒿具有较高的木质素含量和氮含量, 这些性状能够使两种植物被家畜采食量减少, 并保证其具有较强的再生能力, 这可能是糙隐子草和冷蒿耐牧的原因; 大针茅具有最高的植株高度、最大的叶片干物质含量, 以及最高的茎、叶纤维素含量, 说明大针茅是非常典型的竞争物种, 在干扰较低的条件下, 大针茅采取竞争策略对其他物种产生较大的竞争压力可能是其占优势的重要原因。  相似文献

bFGF (basic fibroblast growth factor, bFGF) could promote the proliferation of bone marrow and cord blood mesenchymal stem cells. However, the effect of bFGF on the proliferation of peripheral blood mesenchymal stem cells (PBMSCs) needs further research. This study aimed to investigate the role of bFGF on the culture and expansion of PBMSCs in vitro. Firstly, arterial blood was collected from rats abdominal aorta. After mononuclear cells (MNCs) were separated with Ficoll separation fluid, MNCs were cultured in the DMEM medium without bFGF (served as control group) or with bFGF (10, 20 ng/mL, served as 10 or 20 ng/mL bFGF group). PBMSCs were obtained by adherent culture method. The third passage of PBMSCs was detected for the MSC surface markers and the effect of bFGF on the cell cycle of PBMSCs using flow cytometry. The effects of bFGF on colony formation, cell growth, and the expressions of cyclin D1, cyclin E, p21 and β-catenin were evaluated. PBMSCs showed no difference in morphology among the three groups. PBMSC clonies appeared 14 days after cultivation. Compared with the control group, the cell growth confluence of PBMSCs was obviously increased by 40% and 80% in groups treated with 10 ng/mL bFGF or 20 ng/mL bFGF respectively after culture of 21 days (all P<0.05). Compared with the group treated with 10 ng/mL bFGF, the confluence of PBMSCs in 20 ng/mL group was further increased by 28% (P<0.05). Cells of the third passage were positively stained for CD29 and CD90, while were negative for CD45. These results were consistent with the phenotypic characteristics of MSCs. Compared with the control group, the colony number of PBMSCs in the 10 ng/mL and 20 ng/mL bFGF groups was increased by 51% (P<0.05) and 92% (P<0.05), respectively. Compared with the 10 ng/mL group, the colony number of PBMSCs was further increased in 20 ng/mL group by 14% (P<0.05). The growth curve of PBMSCs showed that after 7 days of culture, the number of PBMSCs in 10 ng/mL bFGF group and 20 ng/mL bFGF group was increased by 41% (P<0.05) and 61% (P<0.05), respectively. Moreover, the cell number had a statistically significant difference between these two groups (P<0.05). Results from flow cytometry cell cycle showed that the numbers of PBMSCs in the G1 phase of experimental groups were significantly decreased as the concentration of bFGF increased when compared with the control group (P<0.05), whereas the number of PBMSCs in the S phase was significantly increased (P<0.05). Immunofluorescence experiments showed that, compared with the control group, bFGF significantly promoted the nuclear translocation and expression of β-catenin in PBMSCs. Compared with the 10 ng/mL group, the PBMSCs in 20 ng/mL bFGF group showed stronger nuclear translocation and expression of β-catenin. Western blot experiments showed that the levels of β-catenin and its target proteins cyclinD1 and cyclinE were significantly increased (all P<0.05), whereas expression of p21 was significantly decreased in PBMSCs in the bFGF groups in a concentration dependent pattern when compared with control group (P<0.05). The study firstly confirms that bFGF promotes the proliferation of PBMSCs by regulating the β-catenin signaling pathway, which may facilitate the aquisition of larger number of PBMSCs for stem cell engineering in vitro.  相似文献

采用溶液培养方法,研究不同浓度硝酸铈对花生(Arachis hypogaea)幼苗生长、开花数目及抗氧化酶过氧化物酶(POD)、超氧化物歧化酶(SOD)活性与丙二醛(MDA)含量的影响。结果表明,与对照相比,铈浓度低于20.0 mg·L–1能促进花生幼苗生长及开花,其中以10.0 mg·L–1铈的效果最为明显,其生物量和开花数分别约为对照的1.3倍和2.8倍;但高于20.0 mg·L–1则抑制花生幼苗生长,降低花朵数目;同时,低于20.0 mg/L铈可抑制花生幼苗过氧化物酶(POD)活性和降低其丙二醛(MDA)含量,其中以10.0 mg·L–1铈的抑制效果最明显,其POD活性和MDA含量约为对照的47.51%和20.76%;而低于20.0 mg·L–1铈能提高花生幼苗的超氧化物歧化酶(SOD)活性,其中以5.0 mg·L–1铈的促进效果最明显,其SOD活性约为对照的2.0倍。  相似文献

Oxidative stress and down-regulated trophic factors are involved in the pathogenesis of nigrostriatal dopamine(DA)rgic neurodegeneration in Parkinson's disease. Fibroblast growth factor 9 (FGF9) is a survival factor for various cell types; however, the effect of FGF9 on DA neurons has not been studied. The antioxidant melatonin protects DA neurons against neurotoxicity. We used MPP+ to induce neuron death in vivo and in vitro and investigated the involvement of FGF9 in MPP+ intoxication and melatonin protection. We found that MPP+ in a dose- and time-dependent manner inhibited FGF9 mRNA and protein expression, and caused death in primary cortical neurons. Treating neurons in the substantia nigra and mesencephalic cell cultures with FGF9 protein inhibited the MPP+-induced cell death of DA neurons. Melatonin co-treatment attenuated MPP+-induced FGF9 down-regulation and DA neuronal apoptosis in vivo and in vitro . Co-treating DA neurons with melatonin and FGF9-neutralizing antibody prevented the protective effect of melatonin. In the absence of MPP+, the treatment of FGF9-neutralizing antibody-induced DA neuronal apoptosis whereas FGF9 protein reduced it indicating that endogenous FGF9 is a survival factor for DA neurons. We conclude that MPP+ down-regulates FGF9 expression to cause DA neuron death and that the prevention of FGF9 down-regulation is involved in melatonin-provided neuroprotection.  相似文献

Synechococcus species are important primary producers in coastal and open‐ocean ecosystems. When nitrate was provided as the sole nitrogen source, nickel starvation inhibited the growth of strains WH8102 and WH7803, while it had little effect on two euryhaline strains, WH5701 and PCC 7002. Nickel was required for the acclimation of Synechococcus WH7803 to low iron and high light. In WH8102 and WH7803, nickel starvation decreased the linear electron transport activity, slowed down QA reoxidation, but increased the connectivity factor between individual photosynthetic units. Under such conditions, the reduction of their intersystem electron transport chains was expected to increase, and their cyclic electron transport around PSI would be favored. Nickel starvation decreased the total superoxide dismutase (SOD) activity of WH8102 and WH7803 by 30% and 15% of the control, respectively. The protein‐bound 63Ni of the oceanic strain WH8102 comigrated with SOD activity on nondenaturing gels and thus provided additional evidence for the existence of active NiSOD in Synechococcus WH8102. In WH7803, it seems likely that nickel starvation affected other metabolic pathways and thus indirectly affected the total SOD activity.  相似文献

ABSTRACT Species reintroductions are used commonly as a tool for conservation, but rigorous, quantitative assessments of their outcome rarely occur. Such assessments are critical for determining success of the reintroduction and for identifying management actions needed to ensure persistence of reintroduced populations. We collected 9 years of demographic data on populations of brown-headed nuthatches (Sitta pusilla) and Eastern bluebirds (Sialia sialis) reintroduced via translocation into Long Pine Key, Everglades National Park, Florida, USA. Realized population growth of brown-headed nuthatches was positive in the first 3 years after cessation of translocations (λ2002 = 1.15, SE = 0.13; λ2003 = 1.28, SE = 0.12; λ2005 = 1.32, SE = 0.20) but became negative thereafter (λ2006 = 0.67, SE = 0.10; λ2007 = 0.77, SE = 0.13). Realized growth rate for the Eastern bluebird population did not vary among years and indicated either a stable or a slowly declining population (λ = 0.92, SE = 0.04). Reintroductions were a qualified success; they resulted in the re-establishment of populations of both species, but neither population grew to the extent expected and both remained at risk of extinction.  相似文献

Comparisons among populations from different localities represent an important tool in the study of evolution. Medflies have colonized many temperate and tropical areas all over the world during the last few centuries. In a common garden environment, we examined whether medfly populations obtained from six global regions [Africa (Kenya), Pacific (Hawaii), Central America (Guatemala), South America (Brazil), Extra-Mediterranean (Portugal) and Mediterranean (Greece)] have evolved different survival and reproductive schedules. Whereas females were either short-lived [life expectancy at eclosion (e0) 48–58 days; Kenya, Hawaii and Guatemala] or long-lived (e0 72–76 days; Greece, Portugal and Brazil], males with one exception (Guatemala) were generally long-lived (e0 106–122 days). Although males universally outlived females in all populations, the longevity gender gap was highly variable (20–58 days). Lifetime fecundity rates were similar among populations. However, large differences were observed in their age-specific reproductive patterns. Short-lived populations mature at earlier ages and allocate more of their resources to reproduction early in life compared with long-lived ones. In all populations, females experienced a post-reproductive lifespan, with this segment being significantly longer in Kenyan flies. Therefore, it seems plausible that medfly populations, inhabiting ecologically diverse habitats, have evolved different life history strategies to cope with local environmental conditions.  © 2009 The Linnean Society of London, Biological Journal of the Linnean Society , 2009, 97 , 106–117.  相似文献

Genome size varies tremendously both within and among taxa, and strong correlations between genome size and various physiological and ecological attributes suggest that genome size is a key trait of organisms, yet the causalities remains vague. In the present study, we tested how genome size is related to key physiological and ecological properties in five large orders of crustaceans: Decapoda, Cladocera, Amphipoda, Calanoida, and Cyclopoida. These span a wide range in sizes, habitats and life-history traits. To some extent, genome size reflected phylogenetic footprints but, generally, a very wide range in genome size was found within all orders. Genome size was positively correlated with body size in Amphipoda, Cladocera, and Copepoda, but not for Decapoda in general. This could indicate that the evolution of body size occurs mainly by changing cell size for the three first orders, whereas it is more attributed to cell numbers for Decapoda. Cladocera, with direct development and a high growth rate, have minute genomes compared to copepods that possess a more complex life history, whereas, within Decapoda and Amphipoda, developmental complexity is not related to genome size. All angiosperms can be arranged along a spectrum from a preponderance of juvenile traits (cambial activity lost) to one of nearly all adult characters (cambium maximally active, mature patterns realized rapidly early in ontogeny). Angiosperms are unique among seed plants in the width of this spectrum. Xylem patterns are considered here to be indicative of contemporary function, not relictual. Nevertheless, most families of early‐divergent angiosperms exhibit paedomorphic xylem structure, a circumstance that is most plausibly explained by the concept that early angiosperms had sympodial growth forms featuring limited accumulation of secondary xylem. Sympodial habits have been retained in various ways not only in early‐divergent angiosperms, but also among eudicots in Ranunculales. The early angiosperm vessel, relatively marginal in conductive abilities, was improved in various ways, with concurrent redesign of parenchyma and fibre systems to enhance conductive, storage and mechanical capabilities. Flexibility in degree of cambial activity and kinds of juvenile/adult expressions has been basic to diversification in eudicots as a whole. Sympodial growth that lacks cambium, such as in monocots, provides advantages by various features, such as organographic compartmentalization of tracheid and vessel types. Woody monopodial eudicots were able to diversify as a result of production of new solutions to embolism prevention and conductive efficiency, particularly in vessel design, but also in parenchyma histology. Criteria for paedomorphosis in wood include slow decrease in length of fusiform cambial initials, predominance of procumbent ray cells and lesser degrees of cambial activity. Retention of ancestral features in primary xylem (the ‘refugium’ effect) is, in effect, a sort of inverse evidence of acceleration of adult patterns in later formed xylem. Xylem heterochrony is analysed not only for all key groups of angiosperms (including monocots), but also for different growth forms, such as lianas, annuals, various types of perennials, rosette trees and stem succulents. Xylary phenomena that potentially could be confused with heterochrony are discussed. Heterochronous xylem features seem at least as important as other often cited factors (pollination biology) because various degrees of paedomorphic xylem are found in so many growth forms that relate in xylary terms to ecological sites. Xylem heterochrony can probably be accessed during evolution by relatively simple gene changes in a wide range of angiosperms and thus represents a current as well as a past source of variation upon which diversification was based.
